I've got an old Marantz PM4200 that I've dug out the loft, planning on putting it to work connected to my iMac along with some old Mission M70's.

The missions are bi-ampable. Question is that the amp supports two sets of speakers, does this mean that I can connect both to my speakers and run them together to bi-amp?

Managed to find a manual online but doesn't reference bi-amping.

I'm going to hook up the iMac with a 3.5m jack to phono lead - can't think of a better way to do it?

Best you'll get is bi-wiring, not bi-amping. Both sets of speaker terminals will be run from the same amp channels.
