Yes
When you get the 3GS, plug in your 3G and let it back everything up in iTunes. Then when you plug your 3GS in it will ask you if you want to restore it from a backup. Select the backup you want and away you go

Thats what happened last year when I went from 2G to 3G

Oh you might want to make sure iTunes is syncing everything thats on the phone first such as Apps, contacts, calendars etc.

Just beware that iTunes doesn't back up photos so you will need to do that manually. In Windows you should be able to do it via explorer and/or the camera wizard; in Mac you can do it with iPhoto I believe.

IF - and it's a big if - the iPhone 3GS does not come with the latest software then you will not be able to back up straight away. You will have to put on the latest software and then when it is completed right click the iPhone icon and up will pop some options to back up from an already saved back up.
Hopefully all the phones will come with 3.0 but we will see.

Re: is there away to backup all the data on my 3g and then transfer it to my new 3gs

Can I backup my 3G version 2.2.1 in iTunes and the connect my new 3GS using version 3.0 and have a restore work or will I need to update the 3G to 3.0 first?

Thanks!
