MDF isn't the ideal material to screw something heavy into - it's easy to strip out the MDF leaving the screw spinning. The thicker the MDF is (ie the longer the screw can be) the less this is likely to happen.
My best suggestion would be to fix the mount to the MDF before (permanently) fixing the MDF to the timber frame - that way you can bolt the mount through the MDF, using large washers to spread the load a little. It may be best to fix the MDF first, mark & drill the holes, then remove the MDF to bolt the mount on.
Obvioulsy then you can fix the MDF to the frame, fill the screwholes, paint etc, and do everything you need to do before mounting the screen.
